Histology
Histological section preparation involves 7 big steps:
- Sampling and fixation
- Paraffin infiltrating
- Embedding
- Microtome sectionning
- Staining
- Coverslipping
- Section control
Sampling and fixation
Whether they reach the laboratory frozen, or already fixed, the received samples are cut according to the protocol that has been defined in the framework of the study. The samples are then placed into cassettes identified by a traceability number and fixated within the adequate safety procedures.
Paraffin infiltrating
An automated process is used to dehydrate and defatten the samples before infiltrating them with paraffin. At HISTALIM, we have several programs depending on the size of samples so as to optimize tissue processing. We have a maximum processing capacity of 1000 histological cassettes per day.
Embedding
The infiltrated tissues are positioned according to the layout defined in the study protocol before being moulded in a paraffin block. In this way, the angle of incidence of the histological sections can be determined and the final images show the required layout.
MICROTOME SECTIONNING
Sectioning quality is an essential point of the histological technique. At HISTALIM, microtome sectioning is entrusted to experienced technicians. Provisions are made in order to ensure the absence of striation, superimposition or contamination of sections.
Staining
Standardized staining protocols at HISTALIM are automated to guarantee the constancy of quality. We determine the shelf life of every dye used, to preserve an irreproachable tinctorial affinity.
Coverslipping
Coverslipping is also automated. It is performed without solvent, i.e. the mounting medium is applied after the evaporation of the solvents to ensure to our customer the excellent preservation of the slides and to prevent the apparition of air bubbles.
Section control
The slides are controlled with a microscope. Control criteria are:
Absence of striation, superimposition or contamination, homogeneous staining intensity, good tinctorial affinity, and absence of micro-bubbles. A document ensuring the traceability of each slide is then edited, validated and joined to the slides, which are dispatched by courier.
